Agentic Nanopore Biosensing

Nanopore transduction detection (NTD) offers to provide an inexpensive, quick, accurate, and versatile method for biosensing, doing assays, glycoassays, PTM-assays. At MLS we are pursuing specific test cases for AI/LLM-managed (real) data analysis. The successful demonstration of Agentic AI signal processing on nanopore data would be hugely significant. Overnight, every nanopore lab or channel current analysis lab, would have the benefit of an AI-based signal processing expert (for free).

A rich assortment of nanopore biosensor applications will become accessible once agentic-level automated AI-based signal processing becomes available. Consider the area of medical diagnostics, for example. It is hypothesized that NTD biomarkers can be developed for early stage disease detection with femtomolar sensitivity, and for doing the standard clinical tests of the future. The NTD approach is also hypothesized to offer a means to observe the behavior (conformational change, etc.) of an individual molecule upon binding. For antibodies, this provides the means to examine their conformational changes upon binding, and improvements of that binding upon introduction of co-factors. NTD methods may, likewise, be used for the selection and refinement of aptamers against a given target. Thus, NTD biomarker biosensing and antibody/aptamer immunoassays are significant capabilities enabled by the NTD approach.

Multi-component and multi-state molecular-conformation regulatory systems and their variations (often sources of disease) could also be studied much more directly using the NTD approach, as could Multi-component (or multi-cofactor) enzyme systems. Information at the single-molecule level may, thus, be uniquely obtainable via NTD methods, and may provide fundamental information regarding kinetic and dynamic characteristics of binding and biomolecular systems critical in biology and medicine.

The operational barrier to a nanopore lab, or channel current analysis effort of any kind, is the in-house signal analysis expertise to process the data available and the new signal analysis demands. To realize the amazing potential of nanopore biosensing, AI Agentic management of the ML-based signal processing tools is needed, and that is what is being pursued at Meta Logos Systems.
